Saphira is a dragon, and as such, she has a much more structured and defined personality than Eragon. She's very focused on achieving her goals and is much less boyish and more serious than Eragon.
I think what makes her character unique and unique is her determination and focus. I know it sounds like an oxymoron, but Saphira is serious and focused on her goals. She's very focused on the mission and the mission only. She's not happy to be there, she's not happy to be with Eragon, and she's not happy to be living at the moment. She is always thinking of how to complete her mission and what she can do to help Eragon succeed.
Eragon is a lot more carefree and much more open to new experiences and ideas. He likes to learn about new things and try new things, but he has a lot of trouble keeping his focus. He can't seem to work on one thing for very long because he wants to do something else.
