Ami is the boss of the 8-bit version of Sonic, and in the Archie comic, she's shown to be a bit more of a tomboy than Amy who is the boss of the modern version of Sonic. Ami also seems to be a bit more aggressive and cocky, which could be because of her being an evil alien.
She's also shown to be somewhat competitive with Amy. The comic shows that a couple of years ago, they were competing against each other to see who was cuter when they were young. Amy won, but it was implied that Ami was still upset about that. This could be why she and Amy get into so many fights throughout the comic, as it's a way for them to compete with each other.
"Ami" is likely short for "Amy", as Amy Rose is named after Amy Wiseman, a character from Sonic the Hedgehog 2. It could also be a tribute to Amy Wiseman, themselves as Amy Rose was based off of her.
